Updating speller matrix

I would like to find out how to update the speller matrix target definitions to define new positions of characters in the grid based on the selected characters. I know I can access the parameter entries e.g. Parameter( "TargetDefinitions" )(x,y). However, I would like to update the entries after character selection.
